The branch will close at 4.00pm on Wednesday, December 4.
The closest branch for customers affected by the closure will be on Horton Street in Port Macquarie.
ANZ general manager for New South Wales and the ACT Amanda Heath-Ogden said there was an "increasing trend across many services to move online for ease of use".
"As the digital economy continues to transform, we are seeing it in our business with many of our customers preferring to use online services," she said.
Ms Heath-Ogden pointed to a survey which found 74 per cent of customers who use Port Macquarie-Oxley as their home branch prefer online and mobile banking compared to 11 per cent who prefer the face-to-face contact the branch provides.
Despite this Ms Heath-Ogden said it wasn't an "easy decision".
"We apologise for the inconvenience this will cause some customers," she said.
"We have written to customers to outline alternative ways to do their banking, including online, telephone and mobile banking.
"Customers who want to continue face-to-face banking can do so at the Port Macquarie branch, which is about 5 kilometres away."
Carol who uses the ANZ branch at Lake Innes told the Port News it was "disappointing".
"I'm sorry it is closing because a lot of the tellers have become friends to us in the last couple of years," she said.
"We now have to go a bit further.
"It was so handy because it was close to our grocery shopping."
Another customer Bernard Hartgill said it was "annoying".
"I only live around the corner and it was very convenient," he said.
"I will now have to go into town.
"I do online banking but I like to do some things face-to-face."
