Port Macquarie-Hastings Council made the decision to close the section of road between Stennetts Road and Hartys Creek Lane after a routine inspection found the deterioration of the road below the surface of an existing major slip.
The road initially reopened to the public in May after the March floods caused the road to sustain extensive damage.
During the closure, Comboyne Community Centre volunteer Grace Gardner said fewer tourists visited Comboyne, as well as the centre.
A detour via Lorne Road, which has unsealed sections, will be in place until further notice for all traffic.
The route will undergo additional maintenance to cater for the increase in traffic flow.
A council spokesperson said council is in the process of engaging a contractor, to complete short term repairs and reopen the Comboyne Road with single lane access.
The work is expected to take three to six weeks and is weather dependent.
Council's director of infrastructure Dan Bylsma said council continues to work on the long term plan for the reconstruction of Comboyne Road.
"Following the flood event earlier this year, council crews sustained an incredible effort to restore the immediate access route for the Comboyne community, while we commenced the process for the longer term planning and design for the major reconstruction of the road," he said.
"Plans for a complete rebuild are already underway and council has begun the procurement process to engage contractors to undertake the longer term project."
"We appreciate this is a key access road for those living in Comboyne and the timing just before Christmas is not ideal. We are working to have the road opened as soon as possible to restore access for the community."
