Online registrations are now open and will be taken from 9.30am on the day. The walk will start at 10am.
This year's walk is the fourth held in Port Macquarie.
The event is the world's biggest type 1 diabetes fundraising event with all funds raised going toward life-changing research.
One of the organisers of Sunday's walk, Emily Acheson says that globally the event will raise over $85m with over one million people participating.
"This success is only possible because of the support, commitment and strength of our community," she said.
"JDRF One Walk is a great way to support the T1D community and medical research. It's a fun family day where T1D and fundraising is the main event.
"T1D is an autoimmune disease in which a person's pancreas loses the ability to produce insulin, a hormone that turns food into energy.

People with T1D require constant blood glucose testing through painful finger pricks, and a constant supply of insulin by injection or pump, just to stay alive.
JDRF Australia CEO Mike Wilson said that last year 20,000 Australians participated in JDRF One Walks around the country, raising more than $1 million for type 1 diabetes research.
"The funds raised at JDRF One Walk are vital to our continued funding for critical type 1 diabetes research," Mr Wilson said.
"A life-long autoimmune disease, type 1 diabetes usually occurs in childhood but can be diagnosed at any age.
"More than 120,000 Australians have this disease, and there are more than seven new diagnoses each day.
"One Walk is a fantastic event that brings together the type 1 community to raise awareness and funds, to help us turn type one into type none."
