Resident Graham Bray said they were extremely concerned about the proposed orbital road.
Concerns include increased traffic, pollution and the loss of a vital bus stop if an orbital road went ahead close to the village.
"The noise levels with traffic revving on four lanes will be high decibels," Mr Bray said.
Hundreds of people live at the Port Macquarie lifestyle village for the over 50s located off Greenmeadows Drive.
The orbital road is proposed as an east-west link (Ocean Drive to Oxley Highway), a north-south link (Oxley Highway to Boundary Street) and a flood-free route to the Port Macquarie Airport.
Port Macquarie-Hastings Council has completed a number of early studies into the potential route options for an orbital road and identified an investigation corridor.
The final route is yet to be determined.
Dahlsford Grove Lifestyle Village Residents' Committee chairman Tom Barnes said residents were concerned about the chance of noise, fumes, pollution and loss of amenity because of increased traffic.
He said it was totally inappropriate for the road to be even considered to come through the Greenmeadows area.
Dahlsford Grove Lifestyle Village resident Bob Sharpham said a lot of people in the village didn't drive and relied on the bus stop right outside the front of the village.
"If they move the bus stop [as a result of the road], it will be too far away for people," he said.
Beth Campbell moved from a Sydney apartment, located near a main road, for a quieter life at the village two and a half years ago.
"One of the main reasons was the noise of the traffic was horrendous and the pollution," she said.
Dahlsford Grove Lifestyle Village director Alan Rosendahl wants the council to consider the impact of the proposed road on people living in the area.
"The level of anxiety in the front of the village is rather significant," he said.
Management has met with mayor Peta Pinson about the proposed orbital road.
Village residents were among those to lodge submissions with the council during the proposed orbital road consultation period.
The council will collate and analyse all the community feedback with an engagement report to be presented to a meeting in June for a decision to determine the project's next stage.
