The highway had been closed in both directions just north of Hastings River Bridge.
Traffic is still heavy in the area, but is expected to return to normal now that all lanes are open.
Motorists should continue to allow additional travel time.
Police are investigating a fatal crash on the Pacific Highway north of Port Macquarie.
Emergency services were called to the Pacific Highway near the intersection of Hastings River Drive at Fernbank Creek, just north of Port Macquarie, shortly after 4pm Friday.
Initial investigations suggest a van and a semi-trailer crashed head on.
The driver of the van, a man, died at the scene.
The passenger of the van, also a man, was trapped for a short time before being released.
He was treated at the scene by Ambulance Paramedics for serious injuries, before being taken to Port Macquarie Hospital.
The driver of the truck was not injured. He will be taken to hospital where he will undergo mandatory blood and urine tests.
The ages of the men involved in the crash are not known at this stage.

Friday 4.30pm: A serious accident north of the Dennis Bridge has closed both lanes of the Pacific Highway.
Motorists have been advised to delay their trips north of Port Macquarie or find alternative routes
Police said the accident was north of the Dennis Bridge.
