The lights were turned on at 10am Tuesday.
Work on the busy intersection began in October as part of the overall upgrade of Hastings River Drive between Park Street and Boundary Street.
The traffic lights coincided with the completion of the Port Macquarie airport upgrade and an expected increase in traffic volume.
The project was undertaken at a cost of approximately $2.8 million and formed part of the adopted operational plan for 2013/14 including a carry-over portion from 2012/13.
Motorists are advised to exercise caution associated with the changed traffic conditions around the new intersection.
Minor works will continue in the area over the next few weeks.
