There will be improved signs around the toilets and waiting area, better access and lighting, and more bike racks.
Seats will be replaced and there will be a new vending machine.
The state government on Tuesday announced the upgrade to the Gordon Street coach terminal which opened in early 2011.
NSW Deputy Premier Troy Grant said the improvements were designed to enhance access and security and encourage more people to take public transport in the region.
"This is a boost for the many customers who use the Port Macquarie interchange, including Mid-North Coast travellers visiting the region," Mr Grant said. He said the funding was a significant investment but he did not divulge the figure so as not to compromise the tender process.
Port Macquarie MP Leslie Williams said trains, buses, taxis and private cars would benefit from the interchange upgrade, with formal taxi and kiss and drop zones making it clearer where drivers could pick up and drop off passengers.
"This upgrade is fantastic news and I know the community will be pleased to see work get underway shortly," she said.
