It is hoped 1000 feet will walk or run along the route from Westport Park to Oxley Beach and back, said Alzheimer's Australia NSW regional manager north coast Gary Thomas.
Registration for walkers remains open on the day and will begin at 7am.
Emcee, Alzheimer's Australia NSW ambassador and actress Doris Younane will officially welcome participants and supporters at 8.10am and the runner and walkers head off from 8.45am.
Mascot for Alzheimer's Australia, Al the Elephant, will meet and greet children and there will be a jumping castle, fun activities and a range of food stalls.
Prizes will be awarded for the best dressed participant and the top fundraisers, plus entertainment and a sausage sizzle operated by Tacking Point Lions Club. The first 500 people to enter will receive a free event shirt.
A contingent of more than 50 staff, volunteers, family and friends from Anglican Care's Taree facilities Storm Village and Bishop Tyrrell Place, will take part in Sunday's walk.
"Anglican Care is very committed to participating in the Memory Walks in the regions in which we operate and we are particularly excited to be participating in the first Walk to be held in Port Macquarie," said Anglican Care marketing and executive support manager Kylie Jacques.
"It is a great way for our staff to contribute to raising funds and awareness for dementia," Ms Jacques said.
Funds will stay in Port Macquarie and go towards supporting the services provided for people living with dementia, their carers, family and friends at the Port Macquarie Dementia and Memory Community Centre.
There are about 1700 people living with dementia in the Port Macquarie region, which is expected to increase to about 2200 by 2020.
