Crime scene and crash investigation officers spent hours on a suburban street after a woman's body was found by the roadside.
It is understood the woman suffered head injuries and died at the scene.
Duty officer Ann Joy said police were called to Hill Street about 6.15pm after a triple-0 call.
Inquiries around the neighbourhood, she said, suggested there may have been a disturbance heard around that time.
While she could not confirm the incident was a hit-and-run, the duty officer said police were keen to speak with anyone travelling through the area at the time.
"It does appear that one neighbour heard a loud noise just prior to quarter past six.
"We need any assistance we can get from members of the general public who may have seen something to contact Port Macquarie police station," duty officer Joy said.
Police have not identified the woman involved but said she was middle-aged with brown shoulder length hair. She was wearing dark clothes and joggers at the time.
