Port Macquarie-Hastings Council group manager infrastructure operations Terry Randall said the Park Street work, between the intersections of Warlters Street and Buller Street, would repair and improve the road surface.
The project will include resurfacing about 120 metres of road over a three-day period.
"We've elected to undertake the work at night, during 21-24 November (subject to weather) between 7pm and 5am, so it can be completed as quickly as possible, while reducing impacts to residents and traffic," he said.
Park Street will be closed each night from 6.30 with detours in place.
Electronic signs by the roadside inform motorists about the project.
