FlyPelican's new service will operate on Thursdays from February 4 with fares starting at $85 one way.
The flights connecting Port Macquarie and Newcastle come after FlyPelican launched its Port Macquarie direct to Canberra service on November 1.
FlyPelican, which is based in Newcastle, flies to a range of destinations.
Newcastle Airport CEO Dr Peter Cock said the continued support of the Newcastle Airport catchment was behind the recent addition of several new services operating to and from Newcastle Airport.
"What we've seen over the last 12 months is strong, ongoing demand for air travel to and from our [Newcastle Airport] catchment which has consistently been higher than the rest of the nation," he said.
"I think it's not only a sign that our regional economy is very resilient but also that people are comfortable using our airport for domestic travel.
"Newcastle Airport offers a stress free, non-congested, Covid-safe environment that gives our customers peace of mind and quick connections to where they want to go."
FlyPelican CEO Marty Hawley echoed Dr Cock's words, saying the continued expansion of his airline's network was a direct result of the ongoing vote of confidence from the Newcastle Airport catchment.
"Despite all the hurdles that the pandemic has thrown up, we've continually found strong demand for inexpensive, convenient travel from our region," he said.
"We're hopeful the addition of this quick flight to Port Macquarie continues that trend."
The flight time will be 30 minutes with one return service a week.
Mayor Peta Pinson was delighted to see Fly Pelican build on the success of the Port Macquarie to Canberra service.
"This is a wonderful new service for our community, providing yet another opportunity for us to spread our wings and travel locally within our beautiful country," Cr Pinson said.
"Local getaways are becoming increasingly popular, and we look forward to welcoming and showcasing our region to even more visitors from Newcastle."
