A group of about 20 will head to the event hoping to gain times to qualify for the Country Championships at the Sydney Olympic Park Aquatic Centre on February 19-21.
Club coach Cheryl Neville said she was confident Port Macquarie had the ability to register some successful results.
"I think they'll go well; a few are training very well," she said.
"Lara Neilson has been training a lot more and she's improving all the time.
"Sally Temple is training a lot and she'll need another year or two to really develop, but her times will be good, I'd love her to get a country time."
Kiera Hetherington is believed to be close to registering national times and Zara Phillips continues to impress.
Taree Torpedoes will host the event at the Manning Aquatic Leisure Centre.
Club secretary Ian Smith said considering spectators, about 1500 people will be in attendance for the two-day event.
"It's a coup not only for the club but for the town. Lots of people are travelling here and they'll need food and accommodation," he said.
