Both teams will take to court six in Port Macquarie at 2.45pm on Saturday, September 26 in the final round of competition hosted by the Hastings Valley Netball Association.
Port Combined have the most recent win (49-41) over the Saints earlier this month on September 12, despite dropping their August 8 match outing (39-27) in favour of the Saints.
Both teams occupy the top two rungs of the ladder with seven wins and two losses.
Port Combined Opens coach Natalie Jordan said many of the players in both teams are friends and there is sure to be some friendly competition over the weekend.
"I think it will be a really, really tight match. Saints have been the benchmark for a number of years now and no one has really been able to get close to them," she said.
"We're really excited to have an opportunity and I think we will give them a good run. We've been focusing on our own game and playing controlled netball.
"Both teams have been at the top for a few years so there's definitely some rivalry there."
Saints captain Jennifer Glover said the team is looking forward to a close match against familiar opponents.
"I think the season so far has gone really well. We have had returning players and new players joining the team," Ms Glover said.
"We're pretty confident but anything can happen so we will just focus to play our best netball. It's always a close contest between us and Port Combined.
"We will stick to our game plan and stick with what works."
