Building and development company Bloc is behind the luxury apartments and townhouses, in a project named Galerie, on vacant land bounded by William Street, Lord Street and Church Street.
Port Macquarie-Hastings Council's Development Assessment Panel granted consent to the development at its meeting on February 12.
The north of the site will be home to an apartment building, with 30 high-end two and three bedroom apartments including two penthouses.
Six large townhouses will front Church Street to the south.
The townhouses and apartments will share driveway access off Lord Street to an 85-space basement car park.
There will be a central courtyard with an indoor pool and spa, alfresco barbecue area and landscaping.
Stewart Architecture is the development's architect.
Bloc director Andrew Redwin said he was really excited about the project.
Bloc has built more than 50 residential apartment buildings between Canberra and Newcastle.
"We are all about building high-end quality apartments, so people can be confident with Bloc," Mr Redwin said.
"We want to build Port Macquarie's premier apartment building - that's our goal."
The exclusive marketing agent for the development is McGrath Estate Agents Port Macquarie.
McGrath Estate Agents Port Macquarie principal Todd Bates said there had been an awesome response to date with about 140 potential buyers registering their interest.
Interest has been received from as far as the US, Melbourne, the Northern Territory and Sydney.
It is expected sales will be launched to the wider market in coming weeks.
"There is a real need for that luxury-type apartment," Mr Bates said.
McGrath Estate Agents Port Macquarie licensed agent and auctioneer Steve Newman said the development would draw lifestyle buyers wanting to downsize to Town Beach retirement living.
He said the Bloc development had perfect timing amid record low interest rates combined with more people moving to town.
Construction will begin after the sales target is met.
