Elders Real Estate sold the two-storey home at 74 The Anchorage for $2.95 million after it went to auction on September 1.
The 877 square metre home boasts five bedrooms and three bathrooms. It features extensive canal views.
The sale price sets the new benchmark with the previous highest price of $2.5m for 4 Major Innes Road and 19 Daintree Lane respectively.
Other high selling properties include an undisclosed price for 108 Hibbard Drive, $2.21m for 73 Matthew Flinders Drive, $2.1m for 4 Grantham Lane, 41 Habourside Crescent and 17 Moondara Terrace and $1.97m for 12 The Peninsula.
Elders Real Estate director David Gray says the new owners have asked not to be disclosed but he confirmed they were from out of the area.
"There were multiple buyers interested in this property, while there were some that could not bid on the day," he said.
"The property was passed in at auction and we were able to negotiate the sale price.
"This sale shows a confidence in the market but it must be remembered that the detail that this owner went to has been rewarded.
"This property is a special home and congratulations to our vendor for their efforts."
Mr Gray says Port Macquarie is experiencing an influx of specialised people to the area with the continued improvements to medical and university facilities.
He said the top end of Port Macquarie's real estate is continuing to generate interest, particularly from the Sydney market.
"I also believe that the lifestyle Port Macquarie has to offer is amazing," he added
Mr Gray said the new record price could also generate further confidence in the market, particularly at the top end.

Considered one of the most prestigious canal addresses, the home is "luxuriously finished in Coastal Hamptons-style", the selling agent says in its description of the home.
Selling agent Corrine Cunningham said the impressive home has set a new precedence for just how strong the growth is in the sought-after coastal town.
