Tacking Point and Aurora Lions clubs, in conjunction with headspace and Marine Rescue, will stage the event which is bigger and better than the successful challenge set competitors in February.
Gaol Point is the launch pad for a field predicted to be up to 1000 people who will head off along the Doctor's Walk to Rocky Point and return to finish at Town Beach.
Along the way there will be 20 obstacles of varying difficulty along the course with some new ones to catch previous entrants out.
There's an ice pit to jump into, major walls to climb and a 15 metre fishing net firmly fixed to the ground which has to be crawled under while other competitors throw buckets of cold water over those trying to negotiate it.
Nowhere will be safe from the sneaky slimers ready to dump gloop over everyone.
A party atmosphere will be greet competitors as they reach the home base with live entertainment by young local DJs Josh Hartmann, Daniel Pye and Ryan Horwood.
Rob Bruce from Tacking Point Lions Club said participants from throughout NSW have been lining up to enter.

The sand mudder will start at 1pm with the last competitors expected home by 5pm.
Music will add to the atmosphere throughout, with the after party starting at 4pm.
Organisers hope the Port Macquarie Sand Mudder will become an annual event attracting large crowds from across the state.
Entrants can be individual competitors, teams of four, fancy dressed, parent/sibling entries and there is a special schools cup event.
"There's even a Zombie wave with a competition for the best Zombie costume," Mr Bruce said.
Entries are open online at www.portmacquariesandmudder.com and information is on Facebook Port Macquarie Sand Mudder.
"This Sand Mudder is going to be bigger, more fun, more diverse, more challenging and have a bigger party than the last event," Mr Bruce said.
All proceeds will stay in Port Macquarie to support headspace, Marine Rescue and Lions.
