Police were called about 9.05pm, with ambulance officers following five minutes later.
A police spokesman said the crash happened because the first car's brakes locked up.
That car was travelling south on Owen Street at "excessive speed" when it braked.
"The brakes harshly locked up the wheels and it collided with a second vehicle that was turning right into Home Street," the spokesman said.
Alcohol was not a factor in the collision, but investigations are continuing.
"There will be infringements issued," the police spokesman said.
An Ambulance NSW spokesman said the two women were taken to Port Macquarie Base Hospital for further treatment.
The women, aged 26 and 30, were in a stable condition on Friday, according to a NSW Health spokeswoman.
Both cars had to be towed from the scene.
An Owen resident said she estimated the first car was speeding at more than 100 kilometres per hour when it braked.
