Running until November 24, classes will start from 10am on Monday mornings and run for two hours with limited spots open for those who were interested.
Council group manager of community place, Lucilla Marshall, said it was a great initiative which would initially only be for seniors.
"It's healthy for elderly people to participate in these sorts of activities because it keeps their minds active," she said.
"It's also another way to get connected in the community and people can try before they buy."
Ms Marshall said they were planning to run another course in February so if people missed out they would have another chance to join in the fun.
"Ukuleles have been getting more and more popular, but this will only be for seniors to start with."
The course is free of charge for seniors and is offered by the Port Macquarie-Hastings Council where people do not have to have any prior musical knowledge to participate.
The course will be run by Port Macquarie Ukestra teacher Ruth Allen and is mainly due to a $4000 Creative Ageing Local Government grant.
A council spokesperson said she wished she was a little bit older so she could take advantage of the free ukulele lessons.
"This is a fantastic initiative for the senior members of our community... who says you're too old to learn something new?" the spokesperson said.
The council will also be the first in Australia which provides ukulele kits for loan during the sessions and the course will take place at the Mac Adams Music Centre (located between Players Theatre and swimming pool).
